Magnesium is an essential mineral and a cofactor for many enzymes. In the nervous system the magnesium ion (Mg2+) blocks the channel of the NMDA-type glutamate receptor in a voltage-dependent way, limiting excitatory signalling, and it has been described as a natural NMDA antagonist and GABA agonist with a role in the regulation of sleep [1]. It is also linked to the hypothalamic-pituitary-adrenal (HPA) axis, the hormonal stress pathway that ends in cortisol release. In a placebo-controlled trial in older adults, 8 weeks of magnesium supplementation raised serum melatonin and renin and lowered serum cortisol compared with placebo [1]. Supplements are sold as different salts (oxide, citrate, glycinate, threonate and others) that differ in the share of elemental magnesium they contain.
